Sequential Immunogold and Cytochemical Stainings for Classification of Acute Lyrnphoblastic Leukemia


Abstract
Immunogold staining in combination with cytochemistry was used to classify acute
lymphoblastic lymphoblastic leukemia by immunological markers.
The monoclonal antibodies OKT3, OKT10, OKIal and the antibody to common ALL
antigen were used for imnlunological marker study and ANAE (alpha-naphthyl acetate
esterase stain), AP (acid phosphatase stain) and PAS (periodic acid schiff's) stain were
used for cytochemical characterization.
The double staining defined clearly both immunological markers and cytochemical
characteristics in each leukemic cell, that made it possible to classify ALL into
common-ALL, T-ALL and B-ALL. ANAE and AP were positive in both OKT10
positive T-ALL and OKT3 positive T-ALL, however PAS positivity did not correlate
with any immunological markers. The percentage of positive cells with immunogold
staining was less than that of immunofluorescence technique.
